Resumo
Introdução: A utilização de produtos de origem vegetal com alto teor de
compostos bioativos mostramse
como importantes elementos na prevenção
do envelhecimento e inovação terapêutica para o reparo tecidual. Objetivo:
Avaliar os efeitos do resveratrol e ácido clorogênico, em marcadores
inflamatórios de linhagem celular estabelecidos de fibroblastos humanos.
Método: Estudo experimental in vitro com fibroblastos CCD1072Sk
cultivados na Unifesp Campus
de São José dos CamposSP.
Foram
realizados testes de viabilidade celular em 24 horas por meio do método de
azul de tripano e para a definição das doses e concentrações da pesquisa foi
utilizado o método MTT. A quantificação do colágeno total foi realizada pelo
método colorimétrico com o corante ácido Sirius Red. Além disso, foram
avaliados os efeitos do Blend na atividade da enzima colagenase, síntese de
ácido hialurônico, expressão de mRNA e secreção das metaloproteinases 1,
2, 9 e interleucina 6. Resultados: O resveratrol e ácido clorogênico,
isoladamente ou na forma de Blend, mostraram efeitos positivos na
promoção da proliferação celular e na modulação da resposta inflamatória,
com redução da expressão de metaloproteinases 1 e da secreção de
interleucina 6 em comparação aos compostos isolados. Conclusão: O
resveratrol e ácido clorogênico de forma isolada e em Blend foram capazes
de modular negativamente os marcadores inflamatórios interleucina 6,
metaloproteinases 1, 2, 9, colagenase e positivamente a síntese de colágeno
e ácido hialurônico na linhagem celular de fibroblastos humanos CCD1072Sk.
Introduction: The use of products of vegetable origin with high content of bioactive compounds are shown as important elements in the prevention of aging and therapeutic innovation for tissue repair. Objective: Evaluate the effects of resveratrol and chlorogenic acid on inflammatory markers in established human fibroblast cell lines. Method: In vitro study with fibroblast line CCD1072Sk was cultivated at Unifesp São José dos Campos CampusSP. Cell viability tests were carried out in 24 hours using the trypan blue method and the MTT was used to define the doses and concentrations of the research. The quantification of total collagen was performed by the colorimetric method with the Sirius Red acid dye. In addition, the effects of the Blend on collagenase enzyme activity, hyaluronic acid synthesis, mRNA expression and secretion of metalloproteinases 1, 2, 9 and IL6 were evaluated. Results: The resveratrol and chlorogenic acid, alone or as a Blend, showed positive effects in promoting cell proliferation and modulating the inflammatory response, with reduced metalloproteinase 1 expression and interleukin 6 secretion compared to the isolated compounds. Conclusion: The resveratrol and chlorogenic acid were able to negatively modulate inflammatory markers interleukin 6, metalloproteinase 1, 2, 9, collagenase and positively modulate collagen and hyaluronic acid synthesis in human fibroblast cell line CCD1072Sk.
