Controle central da atividade física espontânea em camundongos submetidos ou não à restrição calórica durante o processo de envelhecimento

A atividade física espontânea (AFE) engloba toda forma de atividade física, seja em casa, no trabalho ou no lazer. Apesar de negligenciada, ela é um componente importante do balanço energético. Dados anteriores do nosso grupo, relatam um declínio natural da AFE no início da meia idade. Atualmente, a regulação central da AFE é desconhecida, mas existem candidatos para a sua neuro regulação, como é o caso do neuropeptídeo orexina A (OXA). A restrição calórica (RC) retarda as alterações associadas com o envelhecimento, tem efeito neuroprotetor e parece modular positivamente a AFE. Com base no exposto, nosso objetivo foi de verificar se a RC de 30% é capaz de impedir o declínio da AFE durante o envelhecimento e identificar quais áreas do sistema nervoso central (SNC) estão envolvidas. Para tal, camundongos machos C57BL6/J com 4 meses de idade foram distribuídos em grupo controle (alimentado ad libitum até os 10 meses de vida, n =10) e restrito (submetido a RC de 30% até os 10 meses de vida, n=10). A AFE, distância percorrida (DP), velocidade média (VM), porcentagem do tempo em repouso (TR) e a food anticipatory activity (FAA) foram analisados mensalmente, por 24 horas consecutivas. Além disso, parâmetros metabólicos como a evolução da massa corporal, ganho de peso e ingestão alimentar também foram verificados. Ao final do período experimental, os animais realizaram os testes comportamentais de campo aberto e o labirinto em cruz elevado. Após eutanásia, foram analisadas as seguintes áreas do SNC: núcleo accubems (NAcc), núcleo paraventricular do hipotálamo (PVH), núcleo lateral do hipotálamo (LHA), núcleo arqueado do hipotálamo (ARC), substância negra e núcleo dorsal da rafe (DRN). Para entendermos o papel da RC nessas regiões, analisamos os seguintes marcadores, por imunoperoxidase: ΔFosB (atividade neuronal), fator neurotrófico derivado do cérebro (BDNF) e o 8-hidroxideoxiguanosina (8-OHdG, estresse oxidativo). Reconhecendo a importância da OXA na regulação da AFE, sua expressão foi avaliada por imunofluorescência, assim como a sua dupla marcação com o marcador de morte (caspase 3) e sobrevivência celular (Bcl-2) no LHA. A nossa hipótese era que a RC de 30% fosse capaz de reduzir o declínio da AFE durante o envelhecimento e que tal efeito estivesse relacionado com maior atividade neuronal, BDNF e por consequência, uma menor marcação de estresse oxidativo nas áreas encefálicas associadas com a regulação positiva da AFE, incluindo as que expressam OXA. Como resultado, observamos que a RC de 30% imposta durante a vida adulta até o início da meia idade, aumenta consideravelmente a AFE, tanto em quantidade quanto em intensidade. A RC também demonstrou ser capaz de aumentar a DP e diminuir a porcentagem do TR. No entanto, grande parte dessas mudanças aconteceu durante o ciclo claro dos animais, mas sem associações relevantes nos testes comportamentais. Nós também observamos que a FAA é um importante componente da locomoção, mas não foi o único responsável pelo seu aumento nos animais restritos. Em conjunto ao aumento da AFE no protocolo de RC, nós observamos um aumento da atividade neuronal no NAcc e LHA dos animais restritos. Este por sua vez, foi acompanhado também de um aumento dos níveis de BDNF no NAcc, mas com uma diminuição no ARC, nos camundongos submetidos à RC. Além disso, também observamos um aumento da expressão de OXA no LHA dos camundongos submetidos à RC, sem diferenças significativas na dupla marcação de OXA/caspase 3 ou OXA/Bcl-2 no LHA. Não foram observadas diferenças nos níveis de 8OHdG entre os grupos. Como conclusão, acreditamos que a RC de 30% pode ser uma intervenção viável e econômica para aumentarmos a AFE. Além disso, a RC de 30% foi capaz de aumentar a atividade neuronal, o BDNF e a OXA em regiões que são responsáveis pela neuro regulação da AFE. Contudo, ainda é preciso entender melhor como a RC altera o ciclo circadiano e se mesmo quando seu protocolo é interrompido, se os seus benefícios prevalecem.
Spontaneous physical activity (SPA) englobes all forms of physical activity, whether at home, work or leisure. Even neglected, SPA is an important component of the energy balance. Previous data from our group report a natural decline in SPA in early middle age. Currently, the central regulation of SPA is unclear, but there are candidates for its neuro regulation, such as the neuropeptide orexin-A (OXA). In parallel, caloric restriction (CR) delays the changes associated with aging, has a neuroprotective effect and seems to positively modulate the SPA. Based on this scenario, we aimed to verify whether the CR of 30% can prevent the decline in the SPA during aging and to identify which areas of the central nervous system (CNS) are involved. For this purpose, C57bl6/j male mice at 4 months of age were divided into a control group (fed ad libitum until 10 months of age, n =10) and restricted (submitted to CR of 30% until 10 months of age, n = 10). SPA, distance travelled (DT), average speed (AS), resting time (RT) and the food anticipatory activity (FAA) were analyzed monthly, for 24 consecutive hours. In addition, metabolic parameters such as the evolution of body mass, weight gain, and food intake were also verified. In the end, the animals performed the behavioral tests open field and the elevated plus maze. After euthanasia, the following areas of the CNS were analyzed: the nucleus accubems (NAcc), the paraventricular nucleus of the hypothalamus (PVH), the lateral nucleus of the hypothalamus (LHA), the arcuate nucleus of the hypothalamus (ARC), the substantia nigra and the dorsal raphe nucleus (DRN). To understand the role of CR in these regions, we analyzed the following markers by immunoperoxidase: ΔFosB (neuronal activity), brain-derived neurotrophic factor (BDNF), and 8- hydroxydeoxyguanosine (8-OHdG, oxidative stress). Recognizing the importance of OXA in the regulation of SPA, its expression was evaluated by immunofluorescence, as well as its double labeling with the marker of death (caspase 3) and cell survival (Bcl-2) in LHA. We hypothesized that the CR 30% was able to reduce the decline in SPA during aging and that this effect was related to greater neuronal activity, BDNF, and, consequently, a lower marker of oxidative stress in the brain areas associated with the positive regulation of SPA, including those expressing OXA. As a result, we observed that the CR 30% submitted throughout adult life until early middle age considerably increases the SPA levels, both in quantity and intensity. CR has also been shown to be able to increase the DT and decrease the percentage of RT. However, most of these changes took place during the animals' light cycle, but with no relevant associations on behavioral tests. We also observed that FAA is an important component of locomotion, but it was not solely responsible for its increase in CR animals Together with the increase in SPA in the CR protocol, we observed an increase in neuronal activity in the NAcc and LHA of the CR mice. It was also accompanied by an increase in BDNF levels in NAcc, but with a decrease in ARC, in the CR group. In addition, we also observed an increase in OXA expression in the LHA of mice submitted to CR, with no significant differences in the double labeling of OXA/pro caspase and OXA/Bcl-2 in the LHA. No differences in 8OHdG levels were observed between groups. In conclusion, CR may be a viable and low-cost-effective approach to increasing SPA. In addition, CR was able to increase neuronal activity, BDNF, and OXA in the target areas. However, it is still necessary to better understand how CR alters the circadian cycle and whether, even when its protocol is interrupted, its benefits will remain.
