Resumo
Foi conduzido o estudo químico da espécie vegetal Swinglea glutinosa (Rutaceae), uma planta tropical originária do sudeste asiático, caracterizada pela biossíntese de diferentes classes de compostos que possuem diversas atividades biológicas associadas. Para a exploração racional do perfil químico da espécie, utilizou-se a abordagem de desreplicação via cromatografia liquida de alta eficiência (CLAE) acoplada ao espectrômetro de massas de alta resolução (EMAR), bem como dados registrados na literatura, bancos de dados in house e bancos de dados comerciais de Produtos Naturais, além do uso da plataforma GNPS como um facilitador nesse processo. Assim, minimizou-se a chance de isolamento de compostos já descritos na literatura para a espécie. Os extratos e as frações que originaram os metabólitos isolados foram avaliados contra células de leucemia humana (linfoides e mieloides): Jurkat, K-562, Kasumi-1 e Raji. As avaliações preliminares mostraram que algumas frações (na concentração de 100 g/mL) de S. glutinosa inibiram a viabilidade celular de algumas linhagens de leucemia. Com o estudo biomonitorado tentativo e a desreplicação molecular, foi possível isolar quatro compostos inéditos na espécie em estudo: Lansamida I, Lansiumamida B, N-(2-feniletil)cinamamida e Lansiumamida C.
The chemical study of the plant species Swinglea glutinosa (Rutaceae), a tropical plant originating from Southeast Asia, characterized by the biosynthesis of different classes of compounds that have several associated biological activities was conducted. F or the rational exploration of the chemical profile of the species, we used the dereplication approach via high performance liquid chromatography (HPLC) coupled with a high resolution mass spectrometer ( HRMS ), as well as data recorded in the literature, in house databases and commercial databases of Natural Products, in addition to the use of the GNPS platform as a facilitator in this process. Thus, the chance of isolation of compounds already described in th e literature for the species was minimized. The extracts and fractions that originated the isolated metabolites were evaluated against human leukemia cells (lymphoid and myeloid): Jurkat, K 562, Kasumi Preliminary evaluations showed that some e 1 and Raji. xtract fractions (at a concentration of 100 µg/mL) of S. glutinosa inhibited the cell viability of some leukemia strains. With the tentative biomonitored study and molecular dereplication, it was possible to isolate four new compounds in the species under study: Lansamide I, Lansiumamide B, N phenylethyl)cinnamamide and Lansiumamide C.
