dc.description.abstractStable isotopes are becoming increasingly used in research aimed at understanding both the natural dynamics of ecological communities, mapping the diet of a specific animal and analyzing migratory patterns, as well as the general impacts induced by anthropogenic action on ecosystems. The readings are conducted from isotopes of organic molecules, such as carbon and nitrogen present in the keratin of biological systems, for example, of animal hair. However, there is some difficulty in the literature in finding a precise description of the procedures used for the preparation of these samples that will be submitted to reading, leading to the need to develop a protocol that is accessible to laboratories and effective in cleaning and removing other agents that may interfere with the measurement, such as lipids, presented in this document. Elephant seal hair samples were prepared and submitted with six different methodologies for measurements of C and N isotopic ratios in a Delta V Advantage Isotope Ratio Mass Spectrometer (IRMS) coupled to the Isolink Element Analyser (Thermo), and, from the results obtained, comparisons can be made between these values with the different methods proposed, inferring whether there is a significant influence on the values of isotopic ratios due to the presence of lipids in the samples, on which there is still no consensus. Among the methods tested, Groups 3 and 4, which were treated with a solution of Chloroform-Methanol and Extran, presented the lowest and highest nitrogen carbon readings, suggesting that this combination may be the most effective in removing impurities.pt_BR
