Resumo
Introdução: O dimetilaminoetanol (DMAE) tem sido utilizado na prática
clínica no combate a rugas e flacidez cervico-facial. Sua ação firmadora é
explicada devido a sua molécula ser um precursor de acetilcolina, de forma
que o DMAE alteraria a contração muscular. Entretanto não existem
trabalhos experimentais que comprovem esta teoria. Pela falta de definição
do real mecanismo de ação do DMAE, e não existindo referência na
Literatura da sua ação sobre os fibroblastos, foi elaborado estudo para
avaliar a ação direta sobre os fibroblastos humanos cultivados. Métodos:
Fibroblastos humanos provenientes de fragmento de pele total desprezado
de pacientes submetidos a procedimentos cirúrgicos estéticos ou
reparadores realizados na Disciplina de Cirurgia Plástica da UNIFESP/EPM.
Foi utilizada a técnica de explante, e na quarta passagem celular o meio de
cultura foi suplementado com diferentes concentrações de DMAE e as
células foram avaliadas quanto à proliferação, medidas do cálcio citosólico e
ciclo celular. A análise estatística dos resultados foi realizada usando-se o
teste de ANOVA seguido pelo teste de Newman-Keuls para múltiplas
comparações. Resultados: A proliferação dos fibroblastos mostrou-se
diminuída com o aumento das concentrações de DMAE. O tempo de
tratamento com tripsina foi maior nos grupos tratados com DMAE de forma
dose dependente. Houve aumento de cálcio citosólico na presença de
DMAE de forma dose-dependente. Houve um aumento de apoptose nos
grupos tratados com DMAE. Conclusão: O DMAE diminuiu a proliferação
dos fibroblastos, e causou aumento do cálcio citosólico e alterou o ciclo
celular causando um aumento da apoptose nos fibroblastos humanos
cultivados.
Background: Dimethylaminoethanol (DMAE) has been used in medical practice to treat facial wrinkles and cervical flabbiness. Its tensor action is explained by its action as an acethylcoline precursor affecting muscular contraction. However, there are no experimental models proving this theory. Due to the lack of information about DMAE action mechanism and no data about DMAE action on fibroblasts in vitro this study was performed. Methods: Human fibroblasts from patients who undergone cosmetic or reparative surgery performed on Plastic Surgery Division from UNIFESP/EPM were cultured from discharged total skin fragment. Explant technique was used and fibroblasts from passage four were cultured with different concentrations of DMAE. The cells were evaluated in proliferation, cytosolic calcium, and cellular cycle. Statistical analysis was done using ANOVA and Newman-Keuls test for multiple comparisons. Results: Fibroblast proliferation diminished with increasing DMAE concentrations. The trypsin treatment in DMAE groups was longer than in control group in a dose dependent way. Increasing in cytosolic calcium was found in DMAE groups in a dose dependent manner. Apoptosis increased in DMAE treatment group. Conclusion: DMAE diminished fibroblast proliferation, increased cytosolic calcium leading to increasing apoptosis in cultured human fibroblasts.
