Meta-análise do efeito antioxidante e antitumoral do resveratrol em modelos animais com câncer de mama: revisão integrativa para delineamento de estudos clínicos

A utilização da medicina natural tem se tornado cada vez mais frequente, conquistando espaço na indústria de medicamentos e despertando o interesse científico sobre as propriedades medicinais de compostos ativos de origem vegetal. Compostos naturais mostram-se como promissores adjuvantes em diversos tratamentos, com menor índice de efeitos nocivos ao organismo, maior tolerância e conforto para pacientes, principalmente os submetidos a tratamentos mais longos de doenças crônicas. O desequilíbrio do metabolismo do oxigênio pode desencadear a formação de moléculas tóxicas que em excesso geram o estresse oxidativo, aumentando os níveis celulares de radicais livres que comprometem diversas funções biológicas, favorecendo o desenvolvimento de enfermidades degenerativas e/ou crônicas, endócrinas e oncológicas. O câncer é uma das principais doenças crônicas não transmissíveis (DCNT) e de origem multifatorial. O câncer de mama é o tipo de câncer que mais atinge mulheres no mundo, sendo também a causa mais frequente de óbito por câncer entre mulheres. Diversos fármacos quimioterápicos já padronizados para o tratamento oncológico são originados de componentes majoritariamente extraídos de plantas. O resveratrol, principal polifenol da uva, tem sua atividade antioxidante bem estabelecida na literatura, destacando-se o seu potencial efeito antitumoral. O objetivo deste trabalho foi sistematizar de modo quantitativo e qualitativo, dados do efeito do resveratrol na tumorigênese mamária, a partir de estudos in vivo e estudos clínicos. Os resultados obtidos na nossa meta-análise em modelos animais mostraram que o resveratrol apresenta efeito antineoplásico em 85% dos estudos em roedores fêmeas com câncer de mama, atingindo sua maior porcentagem de efeito antitumoral com a dosagem de 25 mg/kg de resveratrol (redução tumoral de 97,8%). Associado com outros antioxidantes, o resveratrol apresentou efeito sinérgico na redução tumoral (86,01%), assim como quando associado a moduladores hormonais (58,33%) e a fármacos antineoplásicos (91%), além de ter sua ação potencializada quando utilizado junto à terapia de radiação (redução tumoral de 89%). Portanto, nossos resultados estatísticos mostraram o resveratrol como potencial agente na prevenção ou adjuvante no tratamento do câncer de mama feminino, em geral amplificando os efeitos antitumorais quando combinado com o tratamento padrão.
The use of natural medicine has become more frequent, conquering space in the medicine industry in order to arouse scientific interest in the medicinal properties of active compounds of vegetal origin. Natural compounds are promising adjuvants in various treatments, with lowest rate of harmful effects to the organism, greater tolerance and convenience for patients, especially those undergoing longer treatments of chronic diseases. The oxygen metabolism imbalance can lead to the formation of toxic products, which in excess generate oxidative stress, increasing cellular levels of free radicals that compromise various biological functions, favoring the development of degenerative and/or chronic, endocrine and oncological diseases. Cancer is one of the main non-communicable chronic diseases (NCDs) and of multifactorial origin. Breast cancer is the type of cancer that most affects women in the world, being the most frequent cause of cancer death between women. Several chemotherapeutic drugs already standardized for cancer treatment originate from components predominantly extracted from plants. The resveratrol, the main grapes polyphenol, has its antioxidant activity well established in the literature, highlighting its potential antitumor effect. The aim of this work was to synthesize, in a quantitative and qualitative means, data on the effect of resveratrol on breast tumorigenesis, based on in vivo and clinical studies. The results obtained in our meta-analysis in animal models showed that resveratrol has an antineoplastic effect in 85% of studies in female rodents with breast cancer, reaching its highest percentage of antitumor effect with a dosage of 25 mg/kg of resveratrol (reduction tumor rate of 97.8%). Associated with other antioxidants, resveratrol had a synergistic effect on tumor reduction (86.01%), as well as when associated with hormonal modulators (58.33%) and antineoplastic drugs (91%), in addition to having its action potentiated when used together with radiation therapy (89% tumor reduction). Therefore, our statistical results showed resveratrol as a potential agent in the prevention or adjuvant treatment of female breast cancer, generally amplifying the antitumor effects when combined with standard treatment.
