- ItemSomente MetadadadosExpressão gênica dos receptores de estrogênios, das ciclinas e dos marcadores de proliferação celular pela genisteína no útero de ratas tratadas imediatamente ou 30 dias após ooforectomia(Universidade Federal de São Paulo (UNIFESP), 2013-09-25) Carbonel, Adriana Aparecida Ferraz [UNIFESP]; Simoes, Manuel de Jesus Simoes [UNIFESP]; Universidade Federal de São Paulo (UNIFESP)Objective: Identification of potential genes of the signaling cascade of estrogen receptors expressed in the uterus of rats treated immediately or lately with genistein or 17β-estradiol. Methods: 60 rats (Rattus norvegicus albinus) females, adult, ± 3 months old were divided into six (6) groups: GI immediate control - animals received vehicle; GII late control - animals received vehicle after 30 days of ovariectomy; GIII immediate genistein - animals treated with genistein (50mg/Kg) 30 days after ovariectomy; GIV late genistein - animals treated with genistein (50mg/Kg) 30 days after ovariectomy; GV immediate estrogen - animals treated with estrogen (17β-estradiol, 10μg/Kg) after ovariectomy and GVI late estrogen - animals treated with estrogen (17β-estradiol, 10μg/Kg) 30 days after ovariectomy. All animals were treated for 30 consecutive days. At the end of treatment, they were anesthetized and part of the uteri removed and immediately immersed in liquid nitrogen for molecular biological analysis and another part in 10% formaldehyde for subsequent histological processing in paraffin for morphological and immunohistochemical study. Statistical analyzes were performed using the software GraphPadPrism ® 5.0 (GraphPad Software Inc.). Statistical test used was one-way ANOVA followed by Tukey test (p < 0,05). Results: Relative expression of ESR2 genes , VEGF - A, MKI67, CCNA1, CCNA2, CCND1, CDK1A, CDKN1B, CDKN1A, CDKNB1 was generally very similar in animals treated immediately or later with genistein (GVI > GV > GIV = GIII > GII = GI), and immediate treatment in the ESR1 gene, showed a higher expression in GIII, however, when compared with GIV was lower than groups GV and GVI . Morphological data showed similar morphology and morphometry in immediate or late treatment. Thus, showing higher endometrial thickness, endometrial surface epithelium, number of glands, glandular circular area, myometrium area and the number of vessels in groups GIV and GV, when compared to other groups. Likewise we observed similar immunohistochemistry results from the beginning of treatment period. As scores of Vegf and Ki67 were higher in GVI (GVI > GV > GIV = GIII > GII = GI). Conclusion: Our data showed that treatment with genistein immediately or 30 days after ovariectomy were similar in relation to the genes of the estrogen receptor pathway, but the immediate treatment have observed increased expression of alpha receptors. The treatment with estrogens obtained greater activation of the gene via the estrogen receptors when administered lately.
