Através da pesquisa descritiva de abordagem qualitativa com egressos do curso de Educação Física. O estudo procurou a partir da experiência construída pelo PET-EF analisar e refletir sobre a relação existente entre universidade (formação acadêmica e produção de conhecimento) e as práticas extensionistas realizadas, quais as contribuições e implicações para o exercício profissional. Os dados foram coletados à partir de entrevistas semiestruturadas e processadas pela análise de conteúdo. O Programa de Educação Tutorial por meio do tripé pesquisa, ensino e principalmente extensão, contribui com a formação e desenvolvimento de características fundamentais para a atuação do profissional de Educação Física, possibilitando o alcance de uma reflexão crítica durante o processo formativo, aprofundada numa concepção de ser humano, de mundo e de sociedade comprometida com práticas sociais éticas na perspectiva da justiça social e da emancipação humana. Dessa forma, conclui-se que o Programa contribui com o processo formativo na graduação e influência na atuação profissional de seus egressos.

While successful treatment with total cure is available, drug resistance threatens patients care in the future and therefore, new antimalarial medicines are required. A potential target for chemotherapeutic intervention is host cell egress, a tightly regulated process that happens after parasites grow and multiply inside the human erythrocytes. Parasite enzymes are known to act in concert dismantling parasitophorous vacuole (PV) and red blood cell (RBC) membranes, but the hierarchy of their action is not completely understood. Objectives: The aims of this study were to establish a method that can be applied to identify compounds that affect egress and to investigate the sequence of the biochemical events involved. Methods: In order to synchronize parasites in two key moments that precede egress, P. falciparum 3D7 parasites were engineered to express endogenous calcium dependent protein kinase 5 (CDPK5) or protein kinase G (PKG) in fusion with a destabilization domain (DD). Expression of each DD tagged kinase is dependent on the ligand Shld-1 and its removal reversibly block egress. Expression of the sensitive reporter nanoluc (NLuc) either secreted do the PV or exported to the RBC was applied to quantify egress. Results: As expected, removal of Shld-1 nearly completely blocked egress in both transgenic lines and when Shld-1 was reintroduced to cultures previously synchronized with sorbitol and heparin at 4548 hours post invasion, Nluc activity in the supernatant increased in CDPK5-DD and PKG-DD cultures after 1 and 2 hours respectively. Microscopic and FACS analysis confirmed the increase in reporter activity correlated to egress. About a couple dozen compounds were evaluated. Egress of CDPK5-DD and PKG-DD were affected differently by a few compounds, demonstrating the method can be applied identify parasite enzymes and/or signalling pathways that act after PKG and before CDPK5. Conclusion: Remarkably, data provided by this study suggest a calpain and a few kinases such as PKA act temporally between PKG and CDPK5. Application of this protocol might allow the identification of many more compounds that affect egress and elucidating when their targets are required respectively to PKG and CDPK5 activation.

The 2001 National Nursing Curriculum Guidelines (DCN-ENF) subsidized the design, construction and content validation of an instrument for the evaluation of graduates of nursing undergraduate courses, known as IAE-Enf. Objectives: Psychometrically validating the IAE-ENF; describing the sociodemographic, economic profile and the insertion of graduates, in an isolated and comparative way in the labor market; comparing the evaluations obtained from the graduates of different institutions in the three dimensions: characterization of the egress, identification and insertion in the labor market, and evaluation of professional training. Method: This is a cross-sectional, multicenter, quantitative methodological study; with data collected between June 2017 and June 2018. The participants (n: 446) are graduates from three undergraduate nursing courses: Federal University of Acre (UFAC), Federal University of São Paulo (UNIFESP) and the State University of Montes Claros (UNIMONTES). Data analysis: IAE-ENF reliability was analyzed by means of test-retest reproducibility with an interval of 7 to 15 days; Cronbach's alpha for internal reliability; exploratory factorial analysis for construct validation using the Kaiser-Meyer-Olkin and Bartlett tests; complemented by Varimax orthogonal rotation and Kaiser's criterion. The essay questions were analyzed according to the content analysis technique and ATLAS ti 8.0 was used. Results: value of the total Cronbach's alpha coefficient of the IAE-ENF was 0.98; the intraclass correlation coefficients of (94%), evidencing strong concordance between the measurements. In the factorial analysis, KMO was 0.971 and the Bartlett sphericity test presented significant results (<0.001) for all constructs and correlation matrices between the items of each construct. The majority subjects were female graduates, between 25 and 30 years old, self-declared of white and ethnic ethnicity (83.37%) and graduates from 2013 to 2016 (80,56%). 54,04% with lato sensu titration completed or in progress. 54,04% with lato sensu titration completed or in progress; 89,27% salaried employees, 60,08% employed in a single institution, in the care area (57,04%), and 59,18% with income ranging from 4 to 7 minimum salaries in force at the time. Dimension 2 of the IAE-ENF denoted high levels of convergence and satisfaction among the graduates from São Paulo and Minas Gerais in the evaluation of learning about activities essential to vocational training, with levels above 50% of agreement. Dimension 3 evidenced a negative evaluation for the group of graduates in the item remuneration, and the essay questions indicated as advantages of the courses the advantages of institutions being public, the quality of teaching staff, teaching, extension and research. As weaknesses, they showed problems with infrastructure, conflicting relationships with the teaching staff, and didactic-pedagogical limitations. Conclusion: the IAE-ENF proved to be valid and reliable for egress evaluation. The items that make up the three dimensions of the IAE-Enf favored the interpretation of the convergence of the DCN-ENF with the academic experience of the graduates and their levels of satisfaction in an isolated and comparative way between the training institutions, denoting the potentialities and fragilities of professional training in Nursing according to geographic region, inclusive.
