Evaluate the microscopic morphology, necrotic area and the collagen content in the rat skin flaps submitted to hyperbaric oxygen therapy (HBOT). Methods. Forty rats adults were divided into four groups: GE ? hair removal; GE / OHB ? hair removal undergone HBO; GER ? hair removal and will perform a skin flap (2 cm wide by 8 cm in length on the back) and GER / OHB ? hair removal, and performed skin flap and submitted to OHB. OHB (2.4 ATA) was performed for two hours, for seven consecutive days. On the eighth day the animals were anesthetized, the flaps removed and separated into three parts: the proximal, middle and distal - with respect to the pedicle fixation. The material was fixed in 10% formalin and processed for paraffin embedding, and the sections stained by HE and picrosirius-red. The blades in HE were examined under a light microscope and collagen content valued at polarizing microscope and ImageLab® software (Bio-Rad). Results. The macroscopic evaluation showed a higher area of necrosis in the three regions of the GER group regarding GE / HBO: (Proximal = 24.6 ± 10.6> 5.6 ± 1.6; Medial; 84.5 ± 5.9> 7.5 ± 3.7; Distal: 98.5 ± 3.7> 15.4 ± 2.5) compared to the GE / OHB. The microscopy study showed a major histopathological lesions in the three regions of the GER group compared to the GE / OHB (Proximal - 8.6 ± 1.2> GER / OHB = 2.4 ± 0.6; medial = 11.5 ± 2. 7> 5.4 ± 1.9 and 14.8 ± 5.3 distal => 8.1 ± 3.7. The collagen concentration was higher in the medial and distal region in GER / OHB group compared to GER (Medial: 81.5 ± 16.7> 55.5 ± 12.6; Distal: 72.2 ± 16.7> 32.8 ± 12.5). Conclusion. Hyperbaric oxygenation had favorable effects on random skin flaps in rats considering the microscopic morphology, necrotic area and collagen concentration.
- ItemSomente MetadadadosEfeitos das experiências adversas precoces e tardias no comportamento alimentar e morfologia da glândula adrenal, fígado e baço de ratos sprague-dawley submetidos ao estresse na vida adulta(Universidade Federal de São Paulo (UNIFESP), 2016-11-29) Pastene, Belgica Katuscha Vasquez [UNIFESP]; Smith, Ricardo Luiz Smith [UNIFESP]; Universidade Federal de São Paulo (UNIFESP)Early interruption of maternal bonding and social isolation are variables involved in social and emotional behavior, which can cause increased anxiety, especially in stressful situations. However, there is no research to explain the phenomenon of overeating anxiety disorder associated with these factors in an animal model and their effects on the morphology of the adrenal gland (AG), liver and spleen. Henceforth, the objective of this research was to determine the effects of early and late intake behavior in adrenal gland, liver and spleen morphology of Sprague-Dawley rats subjected to chronic stress during adulthood. We used 35 newborn female rats, maintained under standardized suckling and feeding conditions. The rats were randomized into 2 control groups (C1 and C2) and 3 experimental groups exposed to an early adverse experience (E1), late (E2) and early-late (E3) subsequently subjected to chronic intermittent stress in adulthood (C2, E1, E2 and E3). Intake behavior of groups and morphological changes of the left AG, the liver and spleen were evaluated by determining morphometric and stereological parameters for each organ. The early and late adverse experiences are factors that influence coping with a stressful situation favoring increased caloric intake and palatable food choice. The morphological features of the AG, the liver and spleen of rats can be affected by exposure to chronic stress, however, when individuals have undergone previous adverse experiences, these changes are more obvious.
- ItemAcesso aberto (Open Access)Histomorphometric analysis of a rat bladder after electrical stimulation(Associação Médica Brasileira, 2011-02-01) Franco, Gisela Rosa [UNIFESP]; Oliveira, Emerson de; Baracat, Edmund Chada [UNIFESP]; Simões, Manuel de Jesus [UNIFESP]; Sartori, Marair Gracio Ferreira [UNIFESP]; Girão, Manoel João Batista Castello [UNIFESP]; Castro, Rodrigo de Aquino [UNIFESP]; Universidade Federal de São Paulo (UNIFESP)OBJECTIVE: We sought to analyze the effects of electrical stimulation (ES) of the pelvic floor on the bladder of rats. METHODS: Forty rats were studied and divided into the following groups: GI (control group) - did not receive ES, GII (placebo) - did not receive ES but had an electrode inserted into the vagina; GIII - underwent six sessions of ES of the pelvic floor and GIV - rats that underwent 12 sessions of ES. Subsequently, the bladder was removed and the epithelium, muscle and blood vessels were analyzed. RESULTS: The muscle wall in GIV had increased thickness when compared to other groups. Further, the number of blood vessels was similar in GIII and GIV, which was higher than that found in GI and GII. Finally, there was an increase in the relative percentage of muscle fibers in relation to collagen for GIV compared to GI. CONCLUSION: After 12 sessions of ES in rats the muscle layer, the number of blood vessels and the relative percentage of muscle fibers were increased.
