Methods: Treatment (56 days) using 32 adult Wistar rats distributed in: group CTRL (saline 0.9% i.p., control diet); Cd (1.2 mg / kg CdCl2 i.p., Control diet); CdJU (1.2 mg / kg CdCl2 i.p., Diet supplemented with 0.5% lyophilized juçara pulp) and JU (0.9% i.p. saline, Diet supplemented with 0.5% lyophilized juçara pulp). The animals were submitted to a single intraperitoneal injection on the 1st day of treatment (90 days of age), and euthanized on the 56th day of treatment (146 days of age). The parameters analyzed were : body mass gain, mass of reproductive organs and accessory glands, daily sperm production, sperm morphology, histopathological and testicular histomorphometric characteristics, liver histopathology and testicular activities of antioxidant enzymes (CAT and SOD). Results: There were no significant diferences related to gain of body mass in either group. The groups Cd and CdJU presented a reduction of the relative mass of the epididymis (p = 0.001), testes (p = 0.001),ventral prostate (p = 0.001) and seminal vesicle (p = 0.003), daily sperm production (p = 0.001), percentage of normal spermatozoa (p = 0.001), reduction in tubular diameter and area (p = 0.001), and a decrease in plasma testosterone levels (p = 0.004). The testes also presented several signs of tissue degeneration (p <0.05). In accordance with the activity of the total and mitochondrial SOD enzyme, there was no significant significant difference between the groups, however, an increase of catalase activity (p = 0.0192) was observed in the animals exposed to cadmium. In all analyzes, the JU group presented no significant difference when compared to the control group. Conclusion: Cadmium was efficient in altering most of the analyzed parameters. The lyophilized juçara pulp, howeever was not able to reverse or attenuate the altered parameters, because its antioxidant content may not have been enough to combat the damage caused by the metal.
- ItemAcesso aberto (Open Access)Estabilidade das antocianinas de juçara: comparação entre os métodos de extração convencional e sistema aquoso bifásico(Universidade Federal de São Paulo, 2018-12-04) Camargo, Marília Ceolin [UNIFESP]; Braga, Anna Rafaela Cavalcante [UNIFESP]; http://lattes.cnpq.br/1145965338858435As antocianinas são polifenóis, pertencentes a classe dos flavonoides, encontrados em plantas superiores, capazes de promover a saúde humana, no entanto, os efeitos das antocianinas em nossa saúde dependem da quantidade consumida e da sua biodisponibilidade. Muitas pesquisas demonstram o potencial antioxidante das antocianinas, além de sua capacidade em combater doenças crônicas neurodegenerativas, cardiovasculares e diabetes. Além das antocianinas serem benéficas a saúde, as mesmas também podem ser empregadas na indústria de cosméticos, farmacêutica e alimentícia como corantes naturais, e na prevenção contra a auto oxidação e peroxidação de lipídeos em sistemas biológicos, entretanto a sua utilização nas industrias ainda é restrita devido à sua baixa estabilidade. Sabe-se que as antocianinas são moléculas muito instáveis físico-quimicamente, e que alguns fatores como a luz, pH, temperatura, presença de oxigênio e de outros íons e moléculas podem causar a sua degradação, além do mais os processos de extração, manipulação, recuperação, purificação e armazenamento também podem influenciar na sua degradação. O estudo da estabilidade térmica das antocianinas obtidas por métodos não convencionais faz-se necessário, a fim de obter métodos mais eficientes e com custo inferior as obtidas pelos métodos convencionais. A polpa dos frutos da palmeira Euterpe edulis Mart., conhecida como juçara, foi escolhida como fonte de antocianinas no desenvolvimento deste estudo, o qual visa comparar a estabilidade térmica das antocianinas extraídas por um método convencional, no qual foi utilizado metanol acidificado, em contraposição ao sistema aquoso bifásico (SAB), em que se empregou polietilenoglicol (PEG)/fosfato de potássio. A partir da análise de degradação térmica foi evidenciado que a estabilidade das antocianinas extraídas utilizando o SAB foi superior, considerando os parâmetros avaliados no presente estudo. A energia térmica de desnaturação e a meia vida, tal como o teor de antocianinas extraídas excedeu àquelas alcançadas pelo método convencional, por conseguinte, o sistema aquoso bifásico apresentou-se como uma excelente alternativa para a extração desse pigmento.

These fruits usually are sources of bioactive compounds, molecules capable of promoting a range of health benefits when consumed regularly by the diet. Among these compounds, the anthocyanins stand out, natural pigments present in several fruits with biological effects that help in the prevention of chronic nontransmissible diseases, and the most studied is the antioxidant activity. However, these properties are considerably reduced due to the exposition of the anthocyanins to variations in factors intrinsic to production processes, such as temperature and light, and to the food digestion process itself, such as pH and oxygen, thus causing changes in the molecular structure of anthocyanins leading to loss of stability. The incorporation of these compounds in nanostructures has been shown to be efficient in maintaining stability and consequent potentiation of antioxidant activity, and the electrospun technique for producing nanostructures is a viable alternative. In this way, the present work aimed to incorporate the pulp of the jussara palm fruit in polyethylene oxide (PEO) nanostructures using the electrospinning technique. From the variation of the process parameters (PEO and NaCl concentrations, flow rate, applied voltage, and collector distance), nanofibers with diameters between 126.00 ± 50 nm and 525.16 ± 120 nm were obtained, and these results directed the process parameters to produce nanostructures containing jussara pulp. Eleven assays were performed using the uniaxial method of electrospinning, and through the response surface methodology, it was found that high concentrations of PEO combined with low concentrations of NaCl, as well as low concentrations of PEO combined with high concentrations of NaCl would result in smaller structures of diameters. For validation of the model obtained, two solutions (7.70% of PEO e 0.36% of NaCl and 8.00% of PEO without salt) were submitted to the electrospinning technique in the uniaxial and coaxial methods. All samples showed low values of diameter measurements, 112.60 ± 45 nm and 118.30 ± 64 nm for the samples obtained by the uniaxial method, and 121.10 ± 54 nm and 110.00 ± 47 nm to the originating from the coaxial process. These results showed the possibility of the use of PEO to incorporate jussara pulp to produce nanofibers. In addition, nanostructures characterization analyzes were performed in order to compare the two methods used. The sample obtained from the coaxial process presents less mass loss and excellent thermal stability. At the same time, FTIR and EDX analyzes proved the efficiency of this method in the maintenance of jussara pulp in the samples.

Studies investigating jussara consumption are least explored at literature, especially under fetal metabolic programming. Objective: To investigate the effect of maternal supplementation with jussara fruit pulp in rats treated with hydrogenated vegetable fat (HVF) during gestation and lactation on the expression of decoupling protein 1 (UCP-1), inflammatory markers in brown adipose tissue (BAT) and mutagenic activity in the liver of 21-day-old of offspring. Methods: The groups were composed of male Wistar rats: Group C - offspring of mothers who received control diet containing soybean oil. Group CJ - offspring of mothers who received C diet with 0.5% supplementation the lyophilized jussara pulp (J). Group T - offspring of mothers who received a diet containing HVF, rich in trans fatty acids (TFAs) and TJ Group - offspring of mothers who received HVF containing diet plus 0.5% of J. Growth and weight gain were evaluated, serum concentrations of HDL-cholesterol, total cholesterol (TC), triglycerides (TG) and glucose, protein contents, the carcass lipid, e TNF-α, IL-10 and IL-6 cytokines, the protein expression of UCP-1, NFκBp50 and RITNF-α in the BAT and the histopathological changes, cytogenetics and immunoreactivity of TNF-α and COX-2 in the liver. To verify the effect the treatment and comparison between the groups, the two-way ANOVA followed by Bonferroni's post hoc test was used. The results were expressed as mean ± standard error of the mean(SEM) and the significance level was p≤0.05. Results: We found that TFAs promoted birth weight gain in first and second week of lactation (TJ> CJ), reduced carcass protein (T C), the concentrations of CT (T> C; TJ> CJ) and TG (T> C) also increased TNF-α (T> C; TJ> CJ), RITNF-α (T> CJ) and UCP-1 (TJ> CJ) in BAT, altered histopathology (T> C), increased the frequency of micronuclei (T> C; T> TJ), immunoexpression to TNF and COX-2 (T> C) in the liver of offspring. In contrast supplementation with jussara reduced the weight gain in the third week of lactation and the total weight gain (CJ T), reduced carcass lipid (CJ T), decreased glycemic (TJ C) and UCP-1 (TJ> T) in BAT, decreased the number of micronucle the hepatocytes (TJ
