Although Studies Show Its Antinociceptive Action, Its Mechanism Of Action Is Still Poorly Understood. Proenkephalin (Penk), Which Is An Endogenous Enkephalin Precursor, Is A Possible Candidate For Bromelain Substrate Due To The Presence Of Pairs Of Basic Amino Acid Residues Flanking The Enkephalin Sequences. Proenkephalin Is Expressed Not Only In The Central Nervous System But Also In The Gastrointestinal Tract And It Is Thus Accessible To Orally Administered Bromelain. The Objective Of This Study Was To Evaluate Whether (1) Hydrolysis Of Proenkephalin By Stem Bromelain In Vivo Is Associated With The Release Of Enkephalin And (2) The Antinociceptive Effect Of Stem Bromelain Depends On Its Proteolytic Activity.
