Estudo de reposicionamento de fármacos potencialmente inibidores da catepsina B1 de Schistosoma mansoni guiado por virtual screening

A esqusitossomose é uma doença negligenciada que coloca cerca de 800 milhões de pessoas em risco de contágio e para a qual o tratamento se apoia em um único fármaco, o praziquantel, que já apresenta relatos de resistência em estudos in vitro e in vivo. Na busca de alternativas terapêuticas, a catepsina B1 do Schistosoma mansoni (SmCB1), cisteíno-protease abundante no estômago deste helminto, mostrou-se alvo interessante por estar intrinsecamente relacionada ao processo de digestão de nutrientes e ao desenvolvimento do parasito e, ainda, por levar à cura parasitológica em camundongos quando inibida, apesar disso, pesquisas relacionadas a inibidores ainda são insipidas. Dentre os métodos de busca de ligantes, o virtual screening (VS) tem se destacado, visto possibilitar a investigação teórica do potencial de ação de conjuntos de moléculas. O VS, ainda, associado a estudos de reposicionamento de fármacos pode levar à economia de tempo e dinheiro. Diante disso, o presente estudo teve como intuito investigar fármacos, por meio de VS, acerca de suas potencialidades como inibidores da SmCB1, empregando a biblioteca do Food and Drug Admnistration (FDA) neste estudo de reposicionamento. Concomitantemente, estudos de docking foram realizados a partir de uma série de 20 análogos sintetizados por Jílková et al. (2011) com vistas a compreender suas relações entre a estrutura química e a atividade biológica (REA). Para realizar o VS farmacóforos foram modelados automaticamente mapeando-se interações entre o cristal 5OGR e o inibidor WRR-286. Estes modelos foram validados utilizando-se curva ROC ao empregar conjuntos de dados ativos e decoys, buscando AUCs superiores a 0,8. Os melhores modelos foram analisados quanto a suas características e capacidade em classificar corretamente instâncias e, então, empregados no VS. Os resultados do VS foram analisados visualmente em termos de acomodação ao sítio enzimático e os ligantes que apresentam sobreposições atômicas (Clashs) foram excluídos das análises futuras. Os melhores ligantes foram analisados quanto ao seu potencial de reposicionamento segundo dados na literatura. Dentre as moléculas selecionadas, a riboflavina (RBF) apresentou os melhores resultados de simulação, sendo a estrutura de maior frequência empregando-se diferentes farmacóforos como referência para o VS. Estudos de docking da RBF apresentaram alta frequência de poses coincidentes, a qual, por sua vez, mostrou-se estável em simulação de dinâmica molecular de 10 ns. Por fim, em estudos in vitro contra S. mansoni, a RBF foi capaz de levar à morte dos vermes bem como à diminuição significativa de motilidade dos parasitos, demonstrando potencial ação esquitossomicida in vitro.
Schistosomiasis is a neglected disease that puts about 800 million people at risk of infection, its treatment is based on a single drug, praziquantel, which has already been reported to be resistant in in vitro and in vivo studies. In the research for therapeutic alternatives, Schistosoma mansoni cathepsin B1 (SmCB1), an abundant cysteine-protease in the stomach of the helminth, proved to be an interesting target because it’s intrinsically related to the process of digestion of nutrients and development of the parasite. Also, because when inhibited it leads to parasitological cure in mice. Yet research related to inhibitors is still scarce. Among the research methods for ligands, virtual screening (VS) has stood out as it enables theoretical investigation on the potential action of sets of molecules. Furthermore, VS, associated to the drug repourposing methodology, can save time and money. Therefore, the present study aimed to investigate drugs, through VS, on their potential as SmCB1 inhibitors, using the Food and Drug Administration (FDA) library on the repourposing study. Concomitantly, docking studies were carried out from a serie of 20 analogues synthesized by Jílková et al. (2011) in order to understand the relationship between chemical structure and biological activity (SAR). To perform the VS, pharmacophores were automatically modeled by mapping interactions between the 5OGR crystal and the WRR-286 inhibitor. These models were validated using the ROC curve using active data sets and decoys, seeking AUCs greater than 0.8. The best models were analyzed for their characteristics and ability to correctly classify instances and then used in VS. VS results were visually analyzed in terms of accommodation to the enzymatic site and ligands that presented clashs were excluded from further analysis. The best ligands were analyzed for their repourposing potential according to literature data. Among the selected molecules, riboflavin (RBF) showed the best simulation results, being the structure with the highest frequency using different pharmacophores as a reference for the VS. RBF docking studies showed a high frequency of coincident poses, which, in turn, was stable in a 10 ns molecular dynamics simulation. Finally, in in vitro studies against S. mansoni, RBF was able to lead worms to death as well as cause a significant decrease in parasite motility, being active as an in vitro as a potencial schistosomicidal agent.
