Objetivo: Estudo da relação entre diferentes temperaturas e períodos de preservação sobre a celularidade de meniscos de coelhos. Métodos: Retirou-se assepticamente 180 meniscos (mediais e laterais) de 45 coelhos (Nova Zelândia), machos, idade entre 5/8 meses, peso médio de 3250g. Os meniscos foram congelados, de dois até trinta dias, a -7,2ºC (n= 60), 21,4ºC (n=60) e –73ºC (n=60). A cada dois dias, de cada temperatura, foram descongelados quatro meniscos e quantificou-se, em quinhentos campos por lâmina, o número de fibroblastos, fibrocondrócitos, condrócitos e picnoses. Resultados: No 14° dia de congelamento a -7,2ºC, a média de células viáveis foi de 92,38 por cento. Entretanto, a partir do 16º dia observou-se uma redução significante de 12 por cento (p=0,001), comparando-se com a média no 14º dia. A média da viabilidade celular, nas temperaturas -21,4ºC e –73ºC, até 16º dia apresentou comportamento estatisticamente semelhante. A partir do 18º dia na temperatura de -21,4ºC a redução do número de células foi significante (p<0,001), especialmente do 28º (54,5 por cento) para o 30º dia (30 por cento) de congelamento. O número de células viáveis a –73ºC mostrou uma redução não significante (p=1,000) de 2,3 por cento no número de células viáveis do 28º (40,2 por cento) ao 30º dia (37,9 por cento). Conclusão: A viabilidade celular dos meniscos congelados diminuiu discretamente até o 14º dia independente da temperatura, e cai rápida e progressivamente a partir de então, sendo mais acentuada nas temperaturas mais baixas de preservação.
Objective: Study of the relationship between different temperatures and rabbits menisci cellularity periods of preservation. Methods: It was removed aseptically 180 medial and lateral menisci of 45 rabbits (New Zelland), male, age between 6/8 months, average weight 3250g. The menisci were frozen, during two until thirty days, at a temperature of -7,2°Celsius (n=60), -21,4°Celsius (n=60) e -73°Celsius (n=60). Four menisci were thawed every two days for histological analysis until completing 30 freezing days, and it was quantified in 500 sield for blade, the number of fibroblasts, fibrochondrocytes and pyknosis. Results: on the 14th defrost day, at the temperature of -7,2°Celsius the average of viable cells was 92,38%. However, after the 16th, it was observed a significant reduction of 12% (p=0,001), comparing with the average of the 14th day. The cells average viability at the temperatures of negative 21,4°Celsius and negative 73°Celsius until the 16th day showed a statistically similar behavior. From the 18th day on at the temperature of -21,4°Celsius the cells number reduction was significant (p<0,001), especially from the 28th (54,5%) to the 30th day (30%) of freezing. The number of viable cells on the temperature -73°Celsius showed no significant reduction (p=1,000) of 2,3% in the from 28th (40,2%) to the 30th day (37,9%). Conclusion: The frozen menisci cells viability was reduced discreetly until the 14th day, independently of the temperature and dropped quickly and progressively and from this day on, being more stressed at lower temperatures.
