- ItemSomente MetadadadosAlterações morfológicas e morfométricas da cérvice uterina determinadas pela ministração local de hialuronidase no final da prenhez da rata albina(Rattus norvegicus albinus, Rodentia, Mammalia)(Universidade Federal de São Paulo (UNIFESP), 2003) Souza, Guilherme Negrão de [UNIFESP]; Camano, Luiz [UNIFESP]Introdução: A validade de uma revisao sistematica e metanalise depende da qualidade do planejamento, realizacao e publicacao dos estudos primarios. Considerando-se a grande variacao existente entre os estudos clinicos randomizados e controlados (ECRC), e importante que se aplique um sistema de avaliacao de qualidade, para que se tenha informacoes sobre a validade do resultado encontrado e da conclusao. Objetivos: O objetivo deste estudo foi analisar se diferentes instrumentos de avaliacao de qualidade, aplicados a um grupo de ECRC, se correlacionam e, se determinam a mesma ordenacao de qualidade destes estudos. Pretendeu-se tambem, testar se a definicao de um escore minimo de qualidade tem impacto na magnitude e/ou direcao do efeito medido na analise. Desenho: Estudo observacional e analitico. Metodo: Foram identificados 38 ECRC sobre a terapeutica da hepatite cronica pelo virus B, com Interferon Alfa, nos principais bancos de dados eletronicos. Aplicou-se aos ECRC os metodos de avaliacao de qualidade de estudos clinicos: Maastricht (M), Delphi (D) e Jadad (J). Duas epidemiologistas clinicas (EC) aplicaram, aos mesmos estudos, o sistema sugerido pela Colaboracao Cochrane, o qual foi assumido como padrao ouro de comparacao. Foi realizado teste de Correlacao de Spearman para se comparar resultados definidos pelos tres instrumentos. O teste de Kappa (K) foi realizado para se avaliar a concordancia entre os revisores, na aplicacao dos instrumentos. Utilizou-se o teste de Kappa Ponderado para se comparar o ordenamento de qualidade determinado pelos instrumentos. Foi realizada metanalise, considerando como desfecho o clareamento do HBV-DNA e HbeAg. Comparou-se os resultados, quando todos os estudos foram incluidos e, quando apenas os considerados melhores o foram, examinando-se o impacto na magnitude e direcao do efeito. Resultados: Em geral, os ECRC foram considerados de regular e baixa qualidade. A concordancia avaliada pelo teste de Kappa, entre os revisores dependeu do instrumento aplicado, situando-se entre pequena e regular (D=0.12; J=0.29 e M=0.33), sendo moderada (0,53) entre as EC. A correlacao entre os instrumentos foi moderada e homogenea (DIJ=0,51; DIM=0,53 e JIM=0,52). Quando todos os artigos identificados foram incluidos na metanalise, (desfecho negativacao do HBV-DNA), obteve-se RR = 0,71; IC 95 por cento: 0,660,77 e, RR = 0,67, 95 por cento: 0,58-0,79, quando so os melhores foram incluidos. Para o desfecho clareamento do HbeAg, os resultados foram respectivamente RR = 0,85; IC 95 por cento: 0,80-0,90 e, RR = 0,85; IC 95 por cento:0,77-0,93. [UNIFESP]; Simoes, Ricardo S.; Baracat, Edmund C.; Soares-, Jose Maria, Jr.The aim of this study was to evaluate the amount of non-and sulfated glycosaminoglycans in the ovariectomized mice uterus, after treatment with ovarian steroids. For this purpose, 50 adult female mice were divided into five groups with 10 animals/each: control group: CG (ovary intact), and ovariectomized groups: OG (vehicle), EG (estradiol), PG (progesterone) and EPG (estradiol combined to progesterone). The treatments started 30 days after ovariectomy. All the animals were treated for 50 consecutive days. These hormones were administered in a sterile oily solution via gavage. Twenty-four hours after the last treatment, all animals were euthanized, removing the uterine horn for biochemical analyses. To quantify, the hyaluronic acid (HA) used ELISA-like fluorometric assay, and the sulfated glycosaminoglycans (GAGs) used agarose gel electrophoresis. The amount of HA was significantly higher in the group treated with progesterone (PG) compared to the others groups (p < 0.05), and in the group treated with estradiol (EG), the amount of chondroitin/dermatan sulfate was significantly higher compared to the others groups (p < 0.05), and in the group treated with progesterone (PG), the amount of heparan sulfate was significantly lower compared to the others groups, except to control group (p < 0.05). Our results showed that the estroprogestative therapy after long time (50 days) profoundly affected the amount of glycosaminoglycans in uterine. These changes may be indicative of uterine pathology such as the development of tumor.

The loss of these compounds facilitates the contact of urine compounds with deeper portions of bladder wall triggering an inflammatory process. We investigated GAG in urine and tissue of PBS/IC and pure stress urinary incontinence (SUI) patients to better understand its metabolism. Materials and Methods: Tissue and urine of 11 patients with PBS/IC according to NIDDK criteria were compared to 11 SUI patients. Tissue samples were analyzed by histological, immunohistochemistry and immunofluorescence methods. Statistical analysis were performed using t Student test and Anova, considering significant when p < 0.05. Results: PBS/IC patients had lower concentration of GAG in urine when compared to SUI (respectively 0.45 ± 0.11 x 0.62 ± 0.13 mg/mg creatinine, p < 0.05). However, there was no reduction of the content of GAG in the urothelium of both groups. Immunofluorescence showed that PBS/IC patients had a stronger staining of TGF-beta, decorin (a proteoglycan of chondroitin/dermatan sulfate), fibronectin and hyaluronic acid. Conclusion: the results suggest that GAG may be related to the ongoing process of inflammation and remodeling of the dysfunctional urothelium that is present in the PBS/IC.

The cellular and molecular knowledge of intervertebral disks composition and precise methods to quantify disk components are important for any type of proposed therapy. Thus, the aim of this study was to correlate glycosaminoglycans presence with the quantitation of cells, ions and collagen fiber distributions in different intervertebral disk sections. Methods: In total, 14 intervertebral disks were used from cattle. All of the disks were dehydrated, separated in seven sections and digested in sodium-free papain buffer. In the majority of accredited training services in Plastic Surgery by the Brazilian Society of Plastic Surgery (SBCP) there are no specific outpatient clinics for attendance and training in Cosmiatry. Objective: To develop a Manual for the implantation of an Ambulatory of Cosmiatry for Accredited Training Services in Plastic Surgery in Brazil. Method: A review of the literature was made on the bases Scielo, Pubmed, Lilacs, selected articles on Cosmiatry. Researched as norms of the National Agency of Sanitary Surveillance (ANVISA) and of the Brazilian Association of Technical Norms (ABNT) for the implantation of medical outpatient clinics. A manual has been created with this data. A questionnaire was applied for method validation using a statistical method - Delphi methodology, together with a group of specialists. Results: Creation of a manual with the selection of contents validated by 7 judges, plastic surgeons with expertise in Cosmiatry, for the implementation of an outpatient clinic for teaching and training in this area. The manual was published in printed and electronic form. Conclusion: A manual was developed for implantation of a Cosmiatry outpatient clinic for Accredited Training Services in Plastic Surgery in Brazil.
